Defining content requirements separately from purity

Turn a reagent quantity requirement into clear procurement fields instead of using a chromatographic purity percentage as a substitute for peptide content.

A quotation can promise high chromatographic purity without answering how much peptide material a supplied amount represents. Before comparing offers, distinguish the quantity being purchased, the content information needed for calculations and the purity criterion used to describe the sample.

Begin with the calculation the study needs

For sourcing, the important consequence is a missing-information question. If the study requires a defined amount of peptide, which part of the quotation establishes that amount, and which additional measurement or definition is needed?

Avoid requesting “the highest purity” as a substitute for answering that question. A more stringent chromatographic specification can still leave the basis of the supplied mass unspecified.

This article addresses how to write the requirement. Selecting a suitable quantitative analytical method requires its own assessment and should not be decided from the wording of a sales description.

Use three independent specification fields

Consider two hypothetical quotations each labelled “10 mg, 98%”. One explicitly states that 10 mg means total powder; the other does not define it. These are not yet comparable offers, even though their headline numbers match.

A useful follow-up asks each supplier to define the 10 mg and 98% separately. It does not assume that multiplying the two gives a valid measured amount of the intended molecular species.

Where a content result is a percentage, request the denominator and any qualification. Where it is an amount per vial, ask whether it describes an individual measurement, a nominal fill or a lot-level assessment.

Distinguish a requested result from a guaranteed minimum

“Please report peptide content” asks for information. “Please supply at least the agreed net amount” asks for a quantity commitment. If both matter, put both in the enquiry instead of expecting either phrase to imply the other.

A supplier may provide content testing as an additional service, as a standard result or not at all. Record the actual response and any limitation; do not describe a test as included because it appears elsewhere in the supplier’s catalogue.

Ask how the quotation will handle a result outside the agreed requirement. Establishing that decision in advance reduces pressure to reinterpret the requirement after the material has arrived.

Compare offers using the same defined quantity

Create a comparison record that preserves unknowns rather than filling them with optimistic assumptions. Price per vial may be clear while price per defined amount remains unavailable.

Keep the supplier’s stated result separate from any calculation performed by the research team. Record the inputs and assumptions so that another reader can see where interpretation begins.

A complete requirement makes it possible to judge whether the delivered information answers the study’s quantity question. It does not turn a purity percentage into a universal guarantee of content, identity or experimental suitability.
